The Interaction of Heat and Lipopolysaccharide on the Expression Levels of Receptor Activator of NF-κB Ligand and Osteoprotegerin in Human Periodontal Ligament Cells

چکیده
Aim: To investigate the expression levels of receptor activator of NF-κB ligand (RANKL) and osteoprotegerin (OPG) in human periodontal ligament fibroblasts when stimulated with heat in infective conditions. Methods: Periodontal ligament fibroblasts were subjected to various temperature increases for 5 min with or without 10 ng/mL lipopolysaccharide (LPS) and then maintained at 37 ̊C for 6 h. After that, the expression levels of RANKL and OPG were investigated using realtime RT-PCR and ELISA. As a positive or negative control, the cells were cultured at 37 ̊C with or without 10 ng/mL LPS. Data were analyzed using one-way ANOVA at a significant level of p = 0.05. Results: The mRNA expression levels of RANKL and OPG were both down-regulated when the cells were heated in infective conditions. The release of sRANKL was increased at low temperatures in such infection; while at high temperatures heat treatment down-regulated the release of sRANKL induced by LPS. The relative RANKL/OPG expression ratios were increased at low temperatures in infective conditions. Conclusions: The interactions between heat and LPS would affect the balance between RANKL and OPG in periodontal ligament fibroblasts when they were heated in infective conditions. Such infection may be the reason why bone resorption occurs in the local area after warm vertical compaction.

Combined Hydroxyapatite Scaffold and Stem Cell from Human Exfoliated Deciduous Teeth Modulating Alveolar Bone Regeneration via Regulating Receptor Activator of Nuclear Factor-Κb and Osteoprotegerin System
Background: Tissue engineering using Stem cell from Human Exfoliated Deciduous Teeth (SHED) and a natural biomaterials biomaterial scaffold has become a promising therapy for the alveolar bone defect. The aim of this study was to analyze the Osteoprotegerin (OPG) and Receptor Activator of NF-Κb ligand (RANKL) expression after the application of Hydroxyapatite scaffold and SHED.Methods: A labora...
متن کاملBacterial PAMPs and Allergens Trigger Increase in [Ca2+]i-induced Cytokine Expression in Human PDL Fibroblasts
An oral environment is constantly exposed to environmental factors and microorganisms. The periodontal ligament (PDL) fibroblasts within this environment are subject to bacterial infection and allergic reaction. However, how these condition affect PDL fibroblasts has yet to be elucidated. PDL fibroblasts were isolated from healthy donors. We examined using reverse transcription-polymerase chain...
متن کاملVGB3 Induces Apoptosis by Inhibiting Phosphorylation of NF-κB p65 at Serine 536 in the Human Umbilical Vein Endothelial Cells
Background and objectives: Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) inhibition results in an increase in apoptosis. It has been demonstrated that NF-κB subunit p65 phosphorylation at the IκB kinase phosphorylation site serine 536 (Ser536) is essential for the NF-κB nuclear translocation and activation. Therefore, NF-κB can be downregulated by suppressing its phosph...

Objective(s): Aberrant expression of CCL5 has been found in several kinds of inflammatory diseases, and the roles of CCL5 in these diseases have also been reported. However, the role of CCL5 in infantile pneumonia is still unclear. Thus, the function and acting mechanism of CCL5 in the in vitro model of infantile pneumonia were researched in this study. Materials and Methods: Human fetal lung f...
